- Abstract
Deoxyribonucleic acid (DNA) extraction is the process of isolating DNA from cells or tissues of living organisms for further analysis. DNA extraction must be performed in matrix analysis of a sample to identify nucleic acids. This study evaluated the effectiveness of ultrasound in extracting pig DNA, with variations in sample type, power, and extraction time as independent variables. The results of the study showed the highest crude DNA pellets in the three samples (processed food, medicine, and cosmetics), which were 40.20 ± 1.10 ng, 0.69 ± 0.18 ng, and 0.10 ± 0.015 ng, respectively. These results were obtained from a combination of high power (450 W), and extraction time (40 min). This indicates that using the ultrasound method with the correct parameters can increase the efficiency of the extraction results through the cavitation mechanism. Ultrasound method is an efficient and environmentally friendly extraction technique for molecular analysis process.
