Advances and Perspectives Chitosan-Sodium Tripolyphosphate as an Effective Drug Delivery System for Skin Applications

- Abstract
Effective topical drug delivery systems pose a significant challenge in dermatological therapy, particularly in overcoming physiological barriers such as stratum corneum penetration, ensuring drug stability, and preventing uncontrolled release. Chitosan–sodium tripolyphosphate (TPP)-based nanoparticles offer an innovative solution, providing advantages in bioadhesion, biocompatibility, encapsulation efficiency, and controlled drug release capabilities. This article reviews recent advances in the formulation, physicochemical characteristics, and mechanism of action of chitosan–TPP nanoparticles in enhancing permeability and therapeutic efficacy. Additionally, this article reviews the potential of this system in delivering various types of active compounds, including those from natural extracts and synthetic drugs. It compares it with other polymer-based nano-particle systems. Advanced formulation approaches, such as ultrasonication after ionic gelation, are also discussed as a strategy to improve particle size and stability of the preparations. Applications include wound healing, skin cancer therapy, treatment of chronic inflammation, and delivery of cosmetic active ingredients. With a multidisciplinary approach and adequate clinical validation, chitosan–TPP nanoparticles have the potential to become a new standard in adaptive, efficient, and personalized dermal drug delivery systems.
