Adverse Event-Based Drug Repurposing for Psoriasis: a Bioinformatics and in Silico Approach

- Abstract
Psoriasis is a chronic autoimmune disease affecting approximately 3% of the global population, with no effective long-term treatment options. This study uses the FAERS database, network pharmacology, and molecular docking to investigate drug repurposing for psoriasis. The FAERS analysis identified ranitidine as a promising candidate, as it showed a negative association with psoriasis in the database. Network pharmacology revealed 13 overlapping genes between psoriasis and ranitidine, which were primarily involved in immune regulation and inflammation. Protein-protein interaction analysis highlighted IL1B as a central gene in the network. Molecular docking simulations further demonstrated that ranitidine interacts with several psoriasis-related proteins, including IL1B, PPARD, and PTPRC, although the binding affinities were generally weaker than those of native ligands. These findings suggest that ranitidine may influence psoriasis-related molecular pathways and support its potential for further experimental validation.
