Abstract

This study primarily used solid dispersion technology to address the problem of Dolutegravir’s poor solubility and rate of dissolution. Using pregelatinized starch as the carrier, solid dispersions were made by physical mixing, kneading, and solvent evaporation techniques. The performance of the resulting formulations was compared. Solvent evaporation technique yielded optimal solid dispersions, demonstrating the highest dissolution rates. These optimized dispersions were then processed into Mouth-Dissolving tablets using direct compression. The resulting tablets (specifically formulation DFT3) met all pharmacopeial standards for stability and physical characteristics, and exhibited excellent drug release, achieving approximately 99% release within 60 minutes, following first-order kinetics. This formulation represents a significant advance toward improving the bioavailability of Dolutegravir.
