Quality of Gaga Chicken Spermatozoa During Freezing with Skim Milk Supplementation in Lactated Ringer’s Diluent

- Abstract
The aim of this study was to assess the quality of Gaga chicken spermatozoa during freezing using lactated ringer diluent added with skim milk. This study used a completely randomized design (CRD) with 4 treatment levels (control, skim milk 5%, 10% and 15%) and 10 replicates. The research stages were diluted spermatozoa, packed in straw, equilibration, pre freezing, freezing and thawing. The parameters observed were plasma membrane integrity and spermatozoa DNA damage. Data were analyzed using analysis of variance (ANOVA), differences between treatments were analyzed using Duncan’s multiple range test. The results showed that there were significant differences (P<0.05) among treatments on plasma membrane integrity and DNA damage before freezing and post thawing and there were no significant differences (P>0.05) among treatments on decreased plasma membrane integrity and increased DNA damage. The use of lactated ringer supplemented with skim milk at a concentration of 5% resulted in better quality of chicken spermatozoa compared to other treatments during freezing.
